What do I provide?

Three ways to use the plasmid DNA preparation service offered at CIAN:

Agar plate(s) with single colonies & liquid growth medium supplemented with appropriate antibiotic (1mL/prep)
OR
Glycerol stocks in the 96-well format
OR
1.5-2mL of overnight cultures of desired clones

When do I submit samples?

Any day before 1:30PM

When can I pick up collection plates?

Next working day at 4:00PM

Same day at 4:00PM

What do I get?

Two sealed 96-well plates containing (i) 30µL/well of plasmid DNA solution and (ii) 10µL/well bacteria culture

One sealed 96-well plate containing 30µL/well of plasmid DNA solution

What are the expected results?

We routinely obtain 3-6µg DNA (100-200ng/µL, with A260/280 ratio between 1.8-2). DNA quality is compatible for sequencing, PCR or restriction digest

Which method is used?

Alkali lysis procedure followed by isopropanol precipitation

Any quality control?

To reduce variability and ensure quality, we use commercial solutions and buffers. In addition, a control is included for each batch of preparation, ensuring quality control

I don’t have 96 samples… Can I use the service?

Yes. Partial plate can be run at the cost of a full plate
